中文摘要: 氨基酰tRNA合成酶是生物体蛋白质合成过程中的一类关键酶 ,对它的研究具有重要的生物学意义和应用前景。该项目系统研究了大肠杆菌亮氨酰 和精氨酰 tRNA合成酶及其与相关tR NA的相互作用。用酶和tRNA的基因克隆和高表达 ,tRNA体外转录 ,基因定点突变 ,酶片段的体外重组等手段研究了酶与tRNA的相互作用的结构基础 ,得到一系列重要结果。

Abstract:Aminoacyl-tRNA synthetases (aaRSs) are a class of essential enzymes in protein biosynthesis. Study on aaRSs is very important to understand the high fidelity during translation and design new antibiotics. In the present research, leucyl- and arginyl-tRNA synthetases from Escherichia coli have been investigated by cloning, over-expression and site-directed mutagenesis of the genes encoding these enzymes and their cognate tRNAs, in vitro trascription, and recombination of peptide fragments. The results from this research work showed many important points to understand the structure basis of accurate interaction between aaRSs and their cognate tRNAs.
